Cypridina luciferase is a secreted luciferase in beautiful blue purple with protein molecular weight of 62KD. Like Gaussia, most of the luciferase products can be secreted into the extracellular area, which can be used for continuous detection of living cells. Because the signal is very strong, the remaining cells in the luciferase is sufficient for routine detection of cell lysis, and multiple detection can be achieved. Cypridina (sea firefly) luciferase and red firefly luciferase can be combined for double color detection.

Red Firefly luciferase is an intracellular protein, whose emission peak is migrated to the red spectral region, compared with the natural Firefly luciferase (green). It is really good news” for luciferase of “flourishing leaves and withering flowers”. It is because of the spectral migration of Red Firefly luciferase, it can separate Gaussia, Cypridina or Green Renilla luciferase to be used in combination.

Beetle luciferase can share the same substrate. They have the same size and most of the amino acids. It is a good choice for dual color detection.

Green Renilla luciferase is an intracellular protein. Compared with the natural Renilla luciferase, it has the higher stability in serum, and the luminescence intensity is better. Therefore, the use of Green Renilla luciferase can get higher reporter gene detection sensitivity.

The same as the ideal reporter gene fluorescent protein, GFP cell now already has a colorful world. Then what are the characteristics of fluorescent protein and luciferase? Fluorescent protein can not get self-luminescence, but requires an excitation light source before it can generate emission fluorescence. Its luminescence energy is from the excited light. The advantages of fluorescent protein do not require substrate and interferential cell for continuous detection. However, the excitatory light can cause background fluorescence.

Single luciferase assay selection is quite rich. Researchers can combine different luciferase for multiple detection according to the needs of study.
