Creative Biostructure, a biotech company specialized in providing contract services to both academia and biotech industries in the field of structural biology, uses both cell-based and cell-free systems to provide customized membrane protein expression services to customers in the pharmaceutical industry, as well as research institutions.

Membrane proteins are involved in many cellular functions, including: signal transduction, cell adhesion, cell movement, transportation, and cell communication. Given its important role in cell function, membrane proteins are often considered to be perfect drug targets. In fact, about two-thirds of the approved drugs target membrane proteins, however, these account for only a small percentage of potential targets. Therefore, membrane proteins are still the main focus of new drug discovery.

Cell-based membrane protein expression
Both prokaryotic and eukaryotic protein expression systems are widely used. Bacterial systems have the advantage of high yield, which is necessary for X-ray crystallography or NMR experiments for structure determination. However, bacteria are not equipped with the complete equipment to complete the required post-translational modifications or correct folding, resulting in protein insolubility or loss of function. Therefore, a variety of eukaryotic cells, such as yeast, insect, plant and mammalian cells, have been developed to facilitate the expression of fully functional proteins.

Cell-free membrane protein expression
In vitro protein synthesis or cell-free protein synthesis (CFPS) is a method of producing proteins in the absence of viable cells but with cell extracts containing the necessary molecular mechanisms. The general cell extract used today is made from E. coli, wheat germ extract, rabbit reticulocyte lysate or insect cell lysate. Production of cell-free proteins can be achieved by adding templates encoding target proteins.

“We have also developed other systems including virus-like particles, liposome, nanodisc, animal-free and detergent-free systems to synthesize soluble and functional membrane proteins. Different systems has its own advantages. Our team will try to choose the most appropriate expression system to obtain high quality membrane proteins to fulfill your specific goals.” commented Joanna from the marketing department at Creative Biostructure.
